Sarah, this is a lovely khamsa!
The letters read "Shaddai" in Hebrew (shin - daled - yud are the Hebrew letters), one of the Jewish mystical names for the Almighty.
On the back of the parchment of mezuzahs, which are hung at the doorpost of (entrance to) Jewish homes, is the Hebrew word 'shaddai '. Shaddai is also an acronym in Hebrew for Shomer Daltot Yisrael, Guardian of the Gates of Israel.
A very nice hamsa~
